Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. <br>
Lane 1: rat liver tissue lysate, <br>
Lane 2: rat spleen tissue lysate, <br>
Lane 3: mouse HEPA1-6 whole cell lysate. <br>
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Lamin B1 antigen affinity purified polyclonal antibody (Catalog # BA1228) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Lamin B1 at approximately 67KD. The expected band size for Lamin B1 is at 67KD.

    背景資料 Lamin-B1 is a protein that in humans is encoded by the LMNB1 gene. The nuclear lamina consists of a two-dimensional matrix of proteins located next to the inner nuclear membrane. The lamin family of proteins make up the matrix and are highly conserved in evolution. During mitosis, the lamina matrix is reversibly disassembled as the lamin proteins are phosphorylated. Lamin proteins are though to be involved in nuclear stability, chromatin structure and gene expression. Vertebrate lamins consist of two types, A and B. This gene encodes one of the two B type proteins, B1.

Instructions

抗體的保存建議:抗體保存得當與否直接決定了抗體的活性和使用效果。如果抗體保存得當,大部分抗體活性都可以維持數月甚至數年。請按照說明書或者抗體標簽上推薦的保存條件正確保存抗體!無論是保存還是運輸,絕對避免反復凍融抗體!

1.拿到抗體后請務必在1000-3000轉離心1-2分鐘后再打開管蓋進行分裝和保存!(由于運輸過程中反復顛簸,管內微量抗體容易聚集在管蓋處,一旦直接打開容易流失抗體。請放心博士德抗體出廠時保證已裝入足量。)

2.對于博士德大部分抗體,比較合適的保存方式是分裝后保存在-20℃冰箱。

◇分裝可以最大程度降低反復凍融對抗體活性的損害,同時也降低了多次從同一管中吸取抗體造成污染的可能性。

◇分裝的量以一次實驗用完為好,但建議最少不要少于10ul每份。分裝體積越小,抗體濃度可能會受到蒸發以及管壁吸附的影響,同時分裝次數越多移液吸頭吸附的抗體也越多,可能會誤認為抗體沒有足量裝入。(最好用無菌的進口0.2ML離心管來分裝抗體,國產的小管一般沒有經過很好的硅化處理,容易吸附抗體蛋白,影響抗體活性。)

◇分裝時請將無菌的微量移液器吸頭先插入管底部反復吹打幾次后再吸出抗體,以便抗體有效成分與抗體保護劑充分混勻。

◇復融后的分裝抗體如果一次用不完,請將剩余母液保存在4℃冰箱,避免再凍起來!

◇絕對避免將抗體保存在自動除霜冰箱的冷藏室中。盡量將抗體保存在手動除霜冰箱里面,而不是在冰箱門上。

3.大部分抗體收到后4℃短暫保存1-2周對抗體活性是基本沒有影響的。如果抗體很快(1-2周內)就能使用完,推薦在4℃保存,避免反復凍融對抗體活性的損害。如果要長期保存則分裝后-20℃保存。

4.即用型抗體請務必保存在4℃冰箱,一定不能冷凍保存。

5.特殊抗體的保存:

◇酶聯抗體:一般保存在4℃,盡量避免冷凍起來。否則可能會導致酶活力的下降或者喪失。

◇偶聯抗體:所有偶聯抗體需要4℃避光保存。尤其是熒光標記抗體,對光極為敏感,因此所有實驗階段也是要避光操作的。
